    Accelerated Metastasis after Short-Term Treatment with a Potent Inhibitor of Tumor Angiogenesis

    SummaryHerein we report that the VEGFR/PDGFR kinase inhibitor sunitinib/SU11248 can accelerate metastatic tumor growth and decrease overall survival in mice receiving short-term therapy in various metastasis assays, including after intravenous injection of tumor cells or after removal of primary orthotopically grown tumors. Acceleration of metastasis was also observed in mice receiving sunitinib prior to intravenous implantation of tumor cells, suggesting possible “metastatic conditioning” in multiple organs. Similar findings with additional VEGF receptor tyrosine kinase inhibitors implicate a class-specific effect for such agents. Importantly, these observations of metastatic acceleration were in contrast to the demonstrable antitumor benefits obtained when the same human breast cancer cells, as well as mouse or human melanoma cells, were grown orthotopically as primary tumors and subjected to identical sunitinib treatments

    Spatio temporal population dynamics of the invasive diatom Didymosphenia geminate in central-southern Chilean rivers

    We document the distribution of Didymosphenia geminata in central-southern Chilean rivers and identify the chemical and physical factors associated with its presence/absence (p/a). Repeated surveys in five successive years provided evidence that D. geminata could be nearing a biogeographic equilibrium in the region. D. geminata databases from extensive biological and environmental surveys in 187 rivers,within ten catchments, south of 38°S commenced in November 2010 and ran through May 2013. In addition, data from two other field surveys were used. The sites evenly distributed latitudinally were climatically characterized. The recent sampling program, following a published species distribution model, was designed to explore D. geminata distribution within thirteen catchments (34°S–48°S). An extensive river survey in 2014 (spring-summer) and in 2015 (autumn) included the p/a, and relative abundance of D. geminata cells in phytobenthos and in the drift. These p/a results showed that the probability of re-encountering D. geminata cells at sites where the species was previously found was significantly highwhile the probability of finding D. geminata cells at sites previously without the species was significantly low. This suggests that the distribution of D. geminata cells among suitable habitats was nearing completion. The relative abundance of D. geminata cells in the phytobenthos versus in the drift indicates seasonality with higher proportion of cells in the phytobenthos during the spring-summer than during the autumn. During the final surveys, principal component analysis of chemical and physical characteristics of rivers showed significant differences between rivers with and without D. geminata. Based on our observations of the distribution of D. geminata cells among rivers with suitable habitat conditions and the fluctuating rate of spread between rivers,we conclude that D. geminata is probably in the ending stage of its spatial demographic expansion in Chile surmounting the different barriers of the invasive process

    Guideline for collection, analysis and presentation of safety data in clinical trials of vaccines in pregnant women.

    Vaccination during pregnancy is increasingly being used as an effective approach for protecting both young infants and their mothers from serious infections. Drawing conclusions from published studies in this area can be difficult because of the inability to compare vaccine trial results across different studies and settings due to the heterogeneity in the definitions of terms used to assess the safety of vaccines in pregnancy and the data collected in such studies. The guidelines proposed in this document have been developed to harmonize safety data collection in all phases of clinical trials of vaccines in pregnant women and apply to data from the mother, fetus and infant. Guidelines on the prioritization of the data to be collected is also provided to allow applicability in various geographic, cultural and resource settings, including high, middle and low-income countries

    Causes of hOCT1-dependent cholangiocarcinoma resistance to sorafenib and sensitization by tumor-selective gene therapy

    Although the multi-tyrosine kinase inhibitor sorafenib is useful in the treatment of several cancers, cholangiocarcinoma (CCA) is refractory to this drug. Among other mechanisms of chemoresistance, impaired uptake through human organic cation transporter type 1 (hOCT1) (gene SLC22A1) has been suggested. Here we have investigated the events accounting for this phenotypic characteristic and have evaluated the interest of selective gene therapy strategies to overcome this limitation. Gene expression and DNA methylation of SLC22A1 were analyzed using intrahepatic (iCCA) and extrahepatic (eCCA) biopsies (Copenhagen and Salamanca cohorts; n = 132) and The Cancer Genome Atlas (TCGA)-CHOL (n = 36). Decreased hOCT1 mRNA correlated with hypermethylation status of the SLC22A1 promoter. Treatment of CCA cells with decitabine (demethylating agent) or butyrate (histone deacetylase inhibitor) restored hOCT1 expression and increased sorafenib uptake. MicroRNAs able to induce hOCT1 mRNA decay were analyzed in paired samples of TCGA-CHOL (n = 9) and Copenhagen (n = 57) cohorts. Consistent up-regulation in tumor tissue was found for miR-141 and miR-330. High proportion of aberrant hOCT1 mRNA splicing in CCA was also seen. Lentiviral-mediated transduction of eCCA (EGI-1 and TFK-1) and iCCA (HuCCT1) cells with hOCT1 enhanced sorafenib uptake and cytotoxic effects. In chemically induced CCA in rats, reduced rOct1 expression was accompanied by impaired sorafenib uptake. In xenograft models of eCCA cells implanted in mouse liver, poor response to sorafenib was observed. However, tumor growth was markedly reduced by cotreatment with sorafenib and adenoviral vectors encoding hOCT1 under the control of the BIRC5 promoter, a gene highly up-regulated in CCA. Conclusion: The reason for impaired hOCT1-mediated sorafenib uptake by CCA is multifactorial. Gene therapy capable of selectively inducing hOCT1 in tumor cells can be considered a potentially useful chemosensitization strategy to improve the response of CCA to sorafenib

    Precision measurements of gamma-ray energies up to 2 MeV using a 2 m curved-crystal monochromator

    Precision measurements of gamma-ray energies up to 2 MeV were performed using a 2m curved-crystal monochromator. Effective source activities of less than 40 mCur were used for the measurements of the transitions in the 2 MeV region. In the 150 keV region, effective source activities of less than 200 [mu]Cur may be used. Measurements have been performed on the transitions of energies less than 2150 keV which follow the decay of In116(54 min), Mn56 (2.6 h) and Sc46 (85 d). In addition, results are presented which indicate that the reflection coefficient is approximately linear with respect to wavelength for the second order reflection from the germanium (02) planes in the region of 6-90 mA (2000-135 keV).Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/31997/1/0000039.pd
